I just got Dynamo today (the simpler version of Reaktor), and the manual is
pretty lame. It explains what each module does, but doesn't really get into
how to use it (I'm assuming that Reaktor's manual does the same).
As far as loading samples, double-click on the name of the sample in the
module window (assuming you're using one of the included modules, it should
preload a sample) -- then you'll get a dialog box to pick your sample. From
there it seems a little confusing with the options to load and replace. I
think I had the best luck when I used replace.
